You can take the Uptown E train (of the A-C-E, the blue line) from 34th Street-Penn Station to 53rd Street-Lexington Avenue. Transfer is available from this station to the 51st Street Station on the 6 train (of the 4-5-6, the dark green line). Walk through the tunnel that connects the two stations, and take the Uptown 6 to 68th Street-Hunter College (at Lexington Avenue).

There's another route that requires less walking, but requires you to go Downtown and then back Uptown: take the Downtown A, C or E from 34th Street-Penn Station to West 4th Street. Transfer at West 4th Street to the Uptown F (of the B-D-F-M, the orange line) and take the F to 63rd Street-Lexington Avenue.

Take the Downtown A-C-E (the blue line) from 34th Street-Penn Station to West 4th Street. Transfer at West 4th Street to the Downtown F train (of the B-D-F-M, the orange line). Take the Downtown F to 2nd Avenue-Houston Street, at 2nd Avenue and East Houston Street.

Get on the Uptown F train (of the B-D-F-M, the orange line) at East Broadway. It's called the East Broadway Station, but the station is actually located on Rutgers Street (which becomes Essex Street at Canal Street, one block north of East Broadway) between Henry Street and East Broadway.

Take the Uptown F from East Broadway to West 4th Street. Then transfer at West 4th Street to the Uptown A-C-E (the blue line) to 34th Street-Penn Station.

You can take the Downtown A-C-E (the blue line) from 34th Street-Penn Station to 14th Street, and transfer there to the Brooklyn-bound L train (the grey line), which runs east along 14th Street.

You can take the L to 14th Street-Union Square (at 4th Avenue/Park Avenue South), 14th Street-3rd Avenue, or 14th Street-1st Avenue.
