When does FMA brotherhood start to differ from the original anime season?

Seeing as Brotherhood is the newer series (or the title of the thirteenth disk in the old one), it is based completely off of the manga series written by Arakawa Hiromu.

Being based off of the books was the original intention of the older series, but there came a point when its creators were forced to deter from the books' plotline. This was because at the time of its creation, the series had no more books from Arakawa-sensei to base their own plotline off of. Hence, the forked pathway.

In the beginning, there are many similarities between the two series, but eventually the older one becomes somewhat hard to believe in its improvised plot twists. I believe it breaks off the path of Arakawa-sensei's intended plot somewhere around the "Psiren Case."

There are a number of characters that appear in Brotherhood, but not in the original series, and vice versa. Lin, and Lanfan are two examples.

Do transmutation circles work?

The origin for transmutation circles came from the anime FullMetal Alchemist. Transmutation circles are formed using the symbols and keys of real life alchemy used during the Middle Ages. Their meanings and the meaning interpreted into the show are very similiar, but that is only done to encourage the belief of it's existence. Creating works that can be linked to our past or everyday lives tends to make us believe in it's existence. So, to answer your question, transmutation circle do not work. They were created to endorse a now famous Japanese anime. and that is all they will ever be.
