Of course! An ukulele with new strings will not hold tuning very well for a week or so, until it settles in. Changes in temperature and humidity can also affect the tuning. You should check the tuning everytime you sit down to play the instrument.

Generally speaking, ukuleles are very low-cost instruments. A low-end Ukulele might cost $20 to $25 dollars, typically sold in bulk to schools for classes for kids. A more durable, well-constructed one can be obtained for around $100. High end models can go into many thousands of dollars for custom made, solid wood and high end components or vintage classic models.

They can be tuned anyway you wish. The most common tuning today is G'CEA, where the C is the lowest note.

Is there a screw on the end of the tuning knob? Tightening it will help hold it in place. Don't over tighten!

If it is a friction peg with no screw at the end, you might try roughing up the pin a bit with some fine sandpaper.

It is possible you would have to replace the tuning pegs. A new set can be obtained for anywhere from $10 to over $100, depending on the quality and style you want. Most of the inexpensive ones will work just fine.
