If you're coming from Manhattan, select either "Grand Central Station" or "Harlem-125th Street" (whichever is more convenient for you) as your "From" station. If you're coming from somewhere else, then, obviously, you would select that location as your "From" station.

You can buy your tickets online ahead of time, OR you can purchase them at the station. You can also buy your tickets after you've boarded the train, but they will be a few dollars more expensive that way.

The two subway lines that stop at Penn Station are the 1-2-3 (the red line) and the A-C-E (the blue line). The 1-2-3 stops at the east side of Penn Station (at 7th Avenue) and the A-C-E stops at the west side (at 8th Avenue).
Both of these subway lines will drop you off very close to 221 Cadman Plaza, so you can just take whichever one you see first.
