The honest answer is that they have no idea, just as they have no idea which chemicals in the food we eat, the 'fresh air' we breathe, the water we drink or the radiation from the sun, mobile phones, etc. cause not only lung cancer but all types of cancer.

They can make an guess from results of epidemiological studies and tests on mice, rats, dogs (try Googling "Beagles Lung Cancer" to see the barbaric cruelty they inflicted to attempt to prove the link between smoking and lung cancer), monkeys, pigs, etc.
