Lauren Faust, the creator of the show, addressed the question of how old the ponies are (see the related link), and said that none of them have defined ages, but rather, have 'maturity levels'. And, as ponies age at a different rate than humans, the Mane 6 could be just 3 or 4 years old and have the maturity level of a young adult human. Lauren Faust has suggested before on her deviantART page that they may be around 4-5 years old as horses mature around age 5, with the CMCs, of course, being younger, possibly 1-3 years old. They almost certainly do not, however, according to her, have 'human-like ages' (e.g. Twilight is more likely 5 than 25), though if you want to see it that way anyways, see the other contributor's answer below.

We can guestimate the age of the Mane 6 (Twilight Sparkle, Apple Jack, Pinkie Pie, Rarity, Rainbow Dash, and Fluttershy) by reasoning. First off, Twilight Sparkle was enrolled into an University at a young age, we can assume maybe 12-14. Now since we don't know much about the school system, this point is pretty much void. However, we can conclude that Twilight Sparkle is a genius from the show's statement that Twilight has the most potential that Celestia has ever seen (Episode 24(?) Cutie Mark Chronicles) So as in any college or University, we can relate Twilight's mission in Ponyville about Friendship is very similar to traveling abroad in our school system. Therefore, we can reason out that Twilight Sparkle is about 18-25, an average age that humans go to college. And since the other Mane 6 ponies all have similar body structures and height to Twilight Sparkle, we can assume they are also around 18-25. The only true fact we know so far about their ages is that Fluttershy is a year older than Pinkie Pie (Episode ? Griffon the Brush Off)
We can also assume, that Mayor Mare, mayor of Ponyville is probably in her late 40's. She has a job that we can assume you keep for life. Also, the art design to give her gray hair and old style looking glasses implies that she is an older pony.
Celestia and Luna are goddesses in this world, so their age is (possibly) infinite, with both sisters being at least 1000 years old.

Recall that Cheerilee was a teenager in the eighties, so she'd be at or around forty now. The Cutie Mark Chronicles shows that Rarity is a year or older, so Rarity must be in her forties.
Also, Mayor Mare's mane is not naturally gray (she dyes it). I figure the Mayor is around fifty or so.
