If you are unwilling to remove all the grout and use a sanded grout, it is best to make sure the grout is cured and sealed properly to limit and minimize cracking.

Its also possible that the floor drain just goes to a dry well which could not be used. Even if it could not be used there are other ways to install a toilet below the septic or sewer line.
