It depends on where in are, and exactly what you mean by the question. In most of the United States, there is no such thing as registration, so the term "registration" is irrelevant in regards to guns. But, to further answer the question, it may not be legal to have a loaded gun that is accessible in your car without a permit depending on what state you are in. In Tennessee for instance, you can only legally carry a loaded gun on your person or have it accessible in your car if you have a permit. You can, however, transport an unloaded pistol without a permit.
