Cylinder misfire can generally occur not only to worn ignition components like the spark plug or wire, though can also be attributed to a faulty fuel injector or a gummed injector. Certain events in a coil-on-plug(COP) design(no wires) if you find your spark plug is known-good or new and gaped properly the coil itself may be faulty/shorted. Looking further, you may have to get further diagnostics to ensure your computer system is sending proper information to the coil(all sensors working in sync).

Depends why the light is on. At my shop I've reset the light with a scan tool and drove through a drive cycle and inspected the vehicle. Unless it's a major problem or a sensor is showing zero reading you can usually put 20-60 highway majority miles and pass. Most vehicles require multiple consecutive failed emission readings before triggering the check engine light.

Open the throttle full open and leave it open for about 5 to 10 minutes, don't try to start it let the excess fuel evaporate. After 10 minutes start it up but don't hit the throttle at all let it start on it's own to burn the excess gas.
I had the same problem and I had to reach away up under the dash and push on the connectors up behind the dash. The hardest to reach ones were not connecting properly and when I pushed and wiggled them it fixed everything.
